DNA filter assay
DNA filter assay A method used to determine the presence of recombinant DNA in cloned cells. This is necessary as a culture will contain many cells that have not been transformed. After the culture has been plated on Petri dishes a copy of the cell pattern is made using a special filter and the cells are then lysed. DNA from the cells binds to the filter as it is heated in a pattern equivalent to that of the cells on the Petri dish. The presence of recombinant DNA is revealed by the use of a gene probe and autoradiography.
