ABSTRACT
Kinetics and voltage dependence of inactivation of a prokaryotic voltage-gated sodium channel (NaChBac) were investigated in an effort to understand its molecular mechanism. NaChBac inactivation kinetics show strong, bell-shaped voltage dependence with characteristic time constants ranging from ~50 ms at depolarized voltages to a maximum of ~100 s at the inactivation midpoint. Activation and inactivation parameters for four different covalently linked tandem dimer or tandem te...

Tryptophan Substitution of a Putative D4S6 Gating Hinge Alters Slow Inactivation in Cardiac Sodium Channels
Biophysical Journal
; ABSTRACT Voltage-gated Na+ channels display rapid activation gating (opening) as well as fast and slow inactivation gating (closing) during depolarization. We substituted residue S1759 (serine), a putative D4S6 gating hinge of human cardiac hNav1.5 Na+ channels with A (alanine), D (aspartate), K

Properties of slow, cumulative sodium channel inactivation in rat hippocampal CA1 pyramidal neurons
Biophysical Journal
; ABSTRACT Sodium channels in the somata and dendrites of hippocampal CAl pyramidal neurons undergo a form of long-lasting, cumulative inactivation that is involved in regulating back-propagating action potential amplitude and can influence dendritic excitation. Using cell-attached patch-pipette

Sodium Channels: Ionic Model of Slow Inactivation and State-Dependent Drug Binding
Biophysical Journal
; ABSTRACT Inactivation is a fundamental property of voltage-gated ion channels. Fast inactivation of Na+ channels involves channel block by the III-IV cytoplasmic interdomain linker. The mechanisms of nonfast types of inactivation (intermediate, slow, and ultraslow) are unclear, although the ionic

Slow Inactivation in Voltage Gated Potassium Channels Is Insensitive to the Binding of Pore Occluding Peptide Toxins
Biophysical Journal
; ABSTRACT Voltage gated potassium channels open and inactivate in response to changes of the voltage across the membrane. After removal of the fast N-type inactivation, voltage gated Shaker K-channels (Shaker-IR) are still able to inactivate through a poorly understood closure of the ion conduction

After 20 years of work, UH researchers think they're close to an answer for heart arrhythmia
Honolulu Star - Bulletin
; Think of the heart as a complex Lego set of little protein machines. Two University of Hawaii researchers have been trying for 20 years to figure out how they work -- or don't work in the case of arrhythmia, an abnormal heart rhythm that potentially can be fatal. "We're down to a very small thing

Tryptophan scanning of D1S6 and D4S6 C-termini in voltage-gated sodium channels
Biophysical Journal
; ABSTRACT Recent reports suggest that four S6 C-termini may jointly close the voltage-gated cation channel at the cytoplasmic side, probably as an inverted teepee structure. In this study we substituted individually a total of 18 residues at D1S6 and D4S6 C-terminal ends of the rNav1.4 Na+ channel

Membrane stretch accelerates activation and slow inactivation in Shaker channels with S3-S4 linker deletions
Biophysical Journal
; ABSTRACT At low P^sub open^(V) Shaker exhibits pronounced stretch-activation. Possible explanations for Shaker's sensitivity to tension include 1) Shaker channels are sufficiently distensible that stretch produces novel channel states and 2) Shaker channels expand in the plane of the membrane

Subunit Dependence of Na Channel Slow Inactivation and Open Channel Block in Cerebellar Neurons
Biophysical Journal
; ABSTRACT Purkinje and cerebellar nuclear neurons both have Na currents with resurgent kinetics. Previous observations, however, suggest that their Na channels differ in their susceptibility to entering long-lived inactivated states. To compare fast inactivation, slow inactivation, and open-channel

Inactivation and pharmacological properties of sqKv1A homotetramers in Xenopus oocytes cannot account for behavior of the squid "delayed rectifier" (K+) conductance
Biophysical Journal
; ABSTRACT Considerable published evidence suggests that a-subunits of the cloned channel sqKv1A compose the "delayed rectifier" in the squid giant axon system, but discrepancies regarding inactivation properties of cloned versus native channels exist. In this paper we define the mechanism of

Inhibition of single Shaker K channels by (kappa)--Conotoxin-PVIIA
Biophysical Journal
; ABSTRACT kappa-Conotoxin-PVIIA (kappa-PVIIA) is a 27-residue basic (+4) peptide from the venom of the predator snail Conus purpurascens. A single kappa-PVIIA molecule interrupts ion conduction by binding to the external mouth of Shaker K channels. The blockade of Shaker by kappa-PVIIA was studied
