ABSTRACT The current study was undertaken to investigate the relative contribution of calcium and myosin binding to thin filament activation. Using the in vitro motility assay, myosin strong binding to the thin filament was controlled by three mechanisms: 1), varying the myosin concentration of the motility surface, and adding either 2), inorganic phosphate (Pi) or 3), adenosine diphosphate (ADP) to the motility solutions. At saturating myosin conditions, Pi had no effect on thin filament mot...

Measurement of thin filament lengths by distributed deconvolution analysis of fluorescence images
Biophysical Journal ; ABSTRACT The lengths of the actin (thin) filaments in sarcomeres directly influence the physiological properties of striated muscle. Although electron microscopy techniques provide the highest precision and accuracy for measuring thin filament lengths, significant obstacles limit their widespread
Role of Ca2+ and cross-bridges in skeletal muscle thin filament activation probed with Ca2+ sensitizers
Biophysical Journal ; ABSTRACT Thin filament regulation of contraction is thought to involve the binding of two activating ligands: Ca^sup 2+^ and strongly bound cross-bridges. The specific cross-bridge states required to promote thin filament activation have not been identified. This study examines the relationship
Fluorescence Depolarization of Actin Filaments in Reconstructed Myofibers: The Effect of S1 or pPDM-S1 on Movements of Distinct Areas of Actin
Biophysical Journal ; ABSTRACT Fluorescence polarization measurements were used to study changes in the orientation and order of different sites on actin monomers within muscle thin filaments during weak or strong binding states with myosin subfragment-1. Ghost muscle fibers were supplemented with actin monomers
Role of Actin C-Terminus in Regulation of Striated Muscle Thin Filament
Biophysical Journal ; ABSTRACT In striated muscle, regulation of actin-myosin interactions depends on a series of conformational changes within the thin filament that result in a shifting of the tropomyosin-troponin complex between distinct locations on actin. The major factors activating the filament are Ca^sup 2+^ and
R-92L and R-92W Mutations in Cardiac Troponin T Lead to Distinct Energetic Phenotypes in Intact Mouse Hearts
Biophysical Journal ; ABSTRACT It is now known that the flexibility of the troponin T (TnT) tail determines thin filament conformation and hence cross-bridge cycling properties, expanding the classic structural role of TnT to a dynamic role regulating sarcomere function. Here, using transgenic mice bearing R-92W and
