Characterization of the BMPR2 5'-Untranslated Region and a Novel Mutation in Pulmonary Hypertension
|
Rationale: Familial pulmonary arterial hypertension results from heterozygous inactivating mutations of the BMPR2 gene. Traditional mutation analysis identifies pathogenic mutations in some 70% of linked families.Wehypothesizedthat the apparent shortfall is due to mutations located in the promoter region of the gene, resulting in abnormal gene regulation.
Objectives: To identify mutations in untranslated sequence regulating BMPR2 transcription.
Methods: DNA upstream of the codin...

Atypical Melting Curve Resulting from Genetic Variation in the 3' Untranslated Region at Position 20218 in the Prothrombin Gene Analyzed with the LightCycler Factor II (Prothrombin) G20210A Assay
Clinical Chemistry
; To the Editor: Carriers of the G20210A allele have high plasma prothrombin concentrations, which in turn are associated with an almost 3-fold increased risk of venous thrombosis (1). As it is the second most common genetic change associated with inherited thrombophilia, the variant site in the 3'

Alignment of the Transcription Start Site Coincides with Increased Transcriptional Activity from the Human Asparagine Synthetase Gene Following Amino Acid Deprivation of HepG2 Cells1
The Journal of Nutrition
; Abstract Transcription initiation sites of the asparagine synthetase gene were investigated in human hepatoma cells after amino acid limitation by incubation in amino acid-complete minimal essential medium or medium lacking histidine. Cells incubated in complete minimal essential medium had mRNA

Structure and hormone responsiveness of the gene encoding the (alpha-Subunit) of the rat amiloride-sensitive epithelial sodium channel
American Journal of Respiratory Cell and Molecular Biology
; The rat amiloride-sensitive epithelial sodium channel (rENaC) is the rate-limiting step for vectorial transport of Na+ across tight epithelia. The complex is composed of three subunits, alpha, beta, and gamma. Expression of the subunits has been shown to be tissue-specific and developmentally and

Core Promoter Sequences Contribute to ovo-B Regulation in the Drosophila melanogaster Germline
Genetics
; ABSTRACT Utilization of tightly linked ovo-A vs. ovo-B germline promoters results in the expression of OVO-A and OVO-B, C^sub 2^H^sub 2^ transcription factors with different N-termini, and different effects on target gene transcription and on female germline development. We show that two

The 5'-untranslated region of the ntp303 gene strongly enhances translation during pollen tube growth, but not during pollen maturation
Plant Physiology
; Transcripts of the ntp303 gene accumulate abundantly throughout pollen development, whereas the protein only accumulates to detectable levels after pollen germination. In an attempt to explain the divergence in the accumulation profiles of the mRNA and the protein, we investigated the role of the

Selenium regulation of classical glutathione peroxidase expression requires the 3' untranslated region in Chinese hamster ovary cells
The Journal of Nutrition
; ABSTRACT Classical glutathione peroxidase (GPX) mRNA levels fall dramatically in selenium (Se)-deficient animals, but it is not known whether this mechanism is related to the mRNA 3' untranslated region (3'UTR) sequences that have been shown to direct Se incorporation. In this study, we used
